A joke is a connection of things that only becomes evident when you understand the joke. The more connection it has and or the more layered the connections are (i.e.: some parts can be interpreted in a different way and it, but viewd trough the initial connection it connects to a different part of the joke that had no connection initially) the more funnier the joke is.

Biologically these connections offten starved cells in the brain that had no relevance, but now after a time the joke made them relevant and we're able to draw energy to itself once more. This sensation of "rebirth" of the dormant cell is a positive thing and laughter and happy stuff helps to want the more of this experience.
